Biological Activity

Chloroquine dihydrochloride is an antimalarial and anti-inflammatory agent that functions as an inhibitor of autophagy and Toll-like receptors (TLRs). It prevents autophagic degradation by impairing lysosomal acidification and blocking fusion between autophagosomes and lysosomes. This disruption of autophagic flux leads to accumulation of autophagic vacuoles and suppresses cellular recycling under stress. Furthermore, inhibition of endosomal TLR pathways disrupts innate immune signaling and suppresses downstream pro-inflammatory cytokine production. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
