Itanapraced (CHF 5074) is a gamma-secretase modulator that displays minimal activity against COX-1 (IC50 > and COX-2 (IC50 >. By selectively modulating gamma-secretase activity, it reduces the secretion of pathogenic amyloid-beta peptides Aβ42 and Aβ40. This shift in amyloid precursor protein processing suppresses microglial activation, decreases brain amyloid plaque deposition, and attenuates neuroinflammation in preclinical models of Alzheimer's disease. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
