Biological Activity

CFT7455 is an orally active degrader of zinc finger transcription factors IKZF1 (Ikaros) and IKZF3 (Aiolos) that binds to cereblon (CRBN) E3 ligase with a Kd of. By recruiting IKZF1 and IKZF3 to the CRL4CRBN E3 ubiquitin ligase complex, CFT7455 promotes their rapid polyubiquitination and proteasomal degradation. This loss of transcription factors disrupts downstream survival pathways, inducing apoptosis and suppressing cell proliferation in multiple myeloma and non-Hodgkin's lymphoma. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
