Cedazuridine (E7727; Compound 7a) is an orally active, synthetic nucleoside analog derived from tetrahydrouridine that functions as a potent cytidine deaminase (CDA) inhibitor. By inhibiting CDA, cedazuridine prevents the rapid deamination and inactivation of cytidine-based nucleoside analogs within the pyrimidine salvage pathway. This enzymatic inhibition preserves target nucleoside exposures, thereby facilitating downstream DNA hypomethylation and epigenetic gene regulation in preclinical models.
