Biological Activity

CCT241533 hydrochloride is an ATP-competitive, selective checkpoint kinase 2 (CHK2) inhibitor with an IC50 of 3 nM. In response to DNA damage, CCT241533 blocks CHK2 signaling pathways by suppressing CHK2 autophosphorylation at S516 and preventing HDMX degradation in human tumor cell lines. Through CHK2 pathway inhibition, it significantly potentiates the cytotoxicity of poly(ADP-ribose) polymerase (PARP) inhibitors in cancer cells. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
