Biological Activity

Mezigdomide (CC-92480) is a cereblon E3 ligase modulator (CELMoD) that functions as a potent protein degrader with high binding affinity for cereblon. By promoting the recruitment of substrate proteins to the CUL4-RBX1-DDB1-CRBN ubiquitin ligase complex, mezigdomide drives the rapid degradation of lymphoid transcription factors Ikaros (IKZF1) and Aiolos (IKZF3). This target degradation suppresses downstream oncogenic signalling and induces apoptosis in multiple myeloma cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
