Biological Activity

Onatasertib (CC 223) is a potent, selective, and orally bioavailable mTOR inhibitor with an IC50 of, exhibiting over 200-fold selectivity for mTOR relative to PI3K-α. By targeting the ATP-binding site of mTOR kinase, it inhibits both mTORC1 and mTORC2 complexes within the AKT/mTOR pathway, effectively suppressing downstream phosphorylation of S6K1 and AKT to impair cancer cell proliferation and survival. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
