CBL0137 is an inhibitor of the histone chaperone FACT (facilitates chromatin transcription) that suppresses NF-κB (EC50 of and activates p53 (EC50 of. By trapping FACT on chromatin, CBL0137 disrupts transcriptional elongation and chromatin remodeling, leading to downstream inhibition of cancer cell proliferation, induction of apoptosis, and suppression of tumor growth across preclinical model systems without inducing direct DNA damage. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
