Biological Activity

CB-5339 is a potent inhibitor of p97, also known as valosin-containing protein (VCP). Selective inhibition of p97/VCP disrupts cellular proteostasis and endoplasmic reticulum-associated degradation, leading to severe accumulation of polyubiquitinated protein substrates. Consequently, this target blockade induces proteotoxic stress, activates the unfolded protein response, and promotes apoptotic cell death while suppressing cancer cell proliferation. [1][2]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
