Biological Activity

CAY10444 (BML-241) is a synthetic thiazolidine carboxylic acid antagonist targeting the sphingosine-1-phosphate 3 (S1P3) receptor. Antagonism of the S1P3 signaling pathway by CAYinhibits ligand-stimulated intracellular calcium mobilization in receptor-expressing cellular models. In addition, CAYsuppresses intracellular calcium elevations driven by purinergic P2 receptors and alpha-1A adrenoceptors, leading to attenuated alpha-1 adrenoceptor-mediated arterial contraction without altering cyclic AMP accumulation. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
