Biological Activity

Calderasib (MK-1084) is a potent and selective macrocyclic covalent inhibitor targeting mutant KRAS G12C. By covalently binding to the switch II pocket of GDP-bound KRAS G12C, it suppresses downstream RAS/RAF/MEK/ERK signaling pathway activation, thereby blocking cancer cell proliferation and suppressing tumor growth. Furthermore, calderasib exhibits robust antitumor efficacy both as a monotherapy and in combination with immune checkpoint inhibitors such as pembrolizumab in KRAS G12C-driven oncology models. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
