BZ1 is a pyridazinone-based inhibitor targeting BPTF with a Kd of 6.3 nM and exhibits greater than 350-fold selectivity over BET bromodomains. BPTF serves as the largest subunit of the nucleosome remodeling factor (NURF) complex to modulate chromatin accessibility. Selective inhibition of BPTF by BZ1 impairs NURF-dependent chromatin remodeling and sensitizes 4T1 breast cancer cells to doxorubicin, highlighting its role in epigenetic regulation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
