BVT 2733 is a non-steroidal, isoform-selective inhibitor of 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1). By blocking 11β-HSD1 activity, it prevents the local conversion of inactive glucocorticoids to active cortisol, thereby downregulating glucocorticoid receptor signaling. This inhibition modulates hepatic metabolic pathways, suppressing gluconeogenic gene expression and enhancing insulin sensitivity to decrease elevated blood glucose levels in metabolic disease models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
