Brigatinib is a potent kinase inhibitor targeting ALK (IC50, 0.6 nM) and ROS1 (IC50, 0.9 nM), while also inhibiting IGF-1R, FLT3, and mutant variants of FLT3 and EGFR with lower potency. By blocking ALK and ROS1 kinase activation, it suppresses downstream STAT3, AKT, and ERK signaling pathways. This inhibition reduces oncogenic signal transduction and inhibits cellular proliferation and survival in driver-dependent cell lines. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
