Biological Activity

BRD4770 is a novel small-molecule inhibitor targeting histone methyltransferase G9a with an IC50 of 6.3 μM. By inhibiting G9a catalytic activity, BRD4770 selectively depletes cellular di-methylated histone H3 lysine 9 (H3K9me2) levels without affecting other methylation states. This blockade of chromatin modification pathways induces downstream cellular senescence, reduces colony formation, and suppresses proliferation in human pancreatic cancer cells.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
