Biological Activity

BRD0639 is a first-in-class, PRMT5 binding motif (PBM)-competitive small-molecule inhibitor that disrupts the interaction between PRMT5 and its substrate adaptor protein RIOK1. By forming a covalent bond with cysteine 278 of PRMT5, BRD0639 inhibits substrate adaptor binding and blocks arginine methylation of target proteins, leading to reduced PBM-dependent PRMT5 methyltransferase activity in cellular models. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
