Biological Activity

Branaplam (LMI070) is a selective small-molecule survival motor neuron-2 (SMN2) splicing modulator with an EC50 of. It promotes the inclusion of exon 7 during SMN2 pre-mRNA splicing, leading to increased production of full-length functional SMN protein. This elevation of SMN protein suppresses motor neuron degeneration and improves phenotypic survival in preclinical models of spinal muscular atrophy.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
