Biological Activity
Boc-Val-Cit-PAB-PNP is a cathepsin B-cleavable dipeptide linker building block utilized in the synthesis of antibody-drug conjugates (ADCs). The valine-citrulline (Val-Cit) motif is specifically recognized and intracellularly cleaved by lysosomal proteases, particularly cathepsin B, following endocytosis of the conjugate. Subsequent self-immolative elimination of the p-aminobenzyl (PAB) spacer releases the active cytotoxic payload inside target tumor cells to induce cell death. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
