5-Boc-amino-pentanoic acid is an alkyl-based PROTAC linker utilized in the chemical synthesis of targeted protein degraders. Incorporating functionalized alkyl linkers enables the conjugation of target-protein ligands to E3 ubiquitin ligase recruiters. Chimeric degraders utilizing such linkers facilitate ubiquitin-proteasome system-mediated degradation of the Polycomb repressive complex 2 core subunits EED, EZH2, and SUZ12, suppressing downstream oncogenic signaling and epigenetic gene repression pathways.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
