BMS-5 is a potent inhibitor of LIM kinase 1 (LIMK1, IC50 = 7 nM) and LIM kinase 2 (LIMK2, IC50 = 8 nM). By inhibiting LIMK activity, BMS-5 blocks downstream cofilin phosphorylation, disrupting actin cytoskeleton remodeling and dynamic turnover. This inhibition leads to severe impairment of actin stress fiber formation, suppressing cell motility, migration, and invasive capacity in cellular models.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
