Biological Activity

BLU9931 is a potent, selective, and irreversible FGFR4 inhibitor displaying selectivity over FGFR1 (297-fold), FGFR2 (184-fold), and FGFR3 (50-fold). By specifically targeting the active site cysteine 552 of FGFR4, it blocks FGF19-driven downstream signaling through the MAPK/ERK pathway, resulting in suppressed cell proliferation and robust antitumor responses in hepatocellular carcinoma models with aberrant FGFR4 activity. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
