Biological Activity

Fisogatinib (BLU-554) is a potent, highly selective, oral FGFR4 inhibitor with an IC50 value of 5 nM, demonstrating marked selectivity over FGFR1-3 (IC50 range. By selectively targeting FGFR4, it blocks FGF19-induced activation of downstream ERK and AKT signaling pathways, leading to suppressed cellular proliferation and induced apoptosis in FGFR4-driven hepatocellular carcinoma models. A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
