BLU-222 is a potent, selective, and orally bioavailable cyclin-dependent kinase 2 (CDK2) inhibitor. It selectively targets the cyclin E/CDK2 complex to block retinoblastoma protein phosphorylation, thereby disrupting G1/S cell cycle progression, inducing p21 and p27 expression, and triggering apoptosis. Through these downstream pathway alterations, BLU-222 suppresses cell proliferation and exhibits robust antitumor activity in CCNE1-aberrant ovarian and endometrial tumors, as well as drug-resistant breast cancers.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
