Biological Activity

BIX-01294 trihydrochloride is a selective inhibitor of G9a histone methyltransferase in a cell-free assay) and weakly inhibits G9a-like protein (GLP). By blocking G9a activity, it reduces histone H3 lysine 9 dimethylation (H3K9me2) in bulk histones without significant activity against other histone methyltransferases, leading to altered gene expression and downstream induction of autophagy. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
