BIBF-0775 acts as a selective inhibitor of transforming growth factor beta receptor I (TGFβRI/ALK5). By target-specific inhibition of ALK5 kinase activity, BIBF-0775 blocks downstream TGF-beta signaling cascades, suppressing the phosphorylation and activation of Smad2 and Smad3 transcription factors. Consequently, suppression of this pathway leads to reduced transcriptional activation of target genes, inhibiting extracellular matrix production, cell proliferation, and epithelial-to-mesenchymal transition in cellular models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
