Biological Activity

BIA is a selective small-molecule antagonist of transmembrane B-cell lymphoma 2-associated X protein inhibitor motif-containing 6 (TMBIM6). By disrupting the specific interaction between TMBIM6 and mTORC2, BIA suppresses mTORC2 assembly and kinase activity. This pathway inhibition blocks downstream AKT phosphorylation and modulates endoplasmic reticulum calcium dynamics, ultimately leading to suppressed cell proliferation, enhanced apoptosis, and inhibited tumor progression in cancer models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
