Bevirimat (PA-457) is a first-in-class HIV-1 maturation inhibitor that targets the Gag polyprotein. It specifically blocks the processing cleavage step at the junction between capsid protein CA and spacer peptide SP1 (CA-SP1) mediated by viral protease. This inhibition disrupts proper viral core condensation and capsid assembly, ultimately leading to the release of immature, non-infectious viral particles and suppressing HIV-1 replication.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
