BCI-121 is a potent inhibitor of the histone lysine methyltransferase SMYD3 that impairs the proliferation of multiple cancer cell lines. By inhibiting SMYD3 activity, BCI-121 suppresses the methylation of downstream signaling substrates such as MAP3K2, leading to attenuated Ras/MEK/ERK pathway activation. As a result, BCI-121 reduces cancer cell growth and survival, making it a valuable tool for investigating SMYD3-dependent oncogenic signaling. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
