BC-1471 is a small-molecule inhibitor of STAM-binding protein (STAMBP) deubiquitinase activity. By inhibiting STAMBP-mediated deubiquitination, this compound decreases NACHT, LRR and PYD domains-containing protein 7 (NALP7) protein levels by promoting endolysosomal degradation. Downstream, STAMBP inhibition suppresses Toll-like receptor (TLR) agonism-induced inflammasome activation and reduces pro-inflammatory interleukin-1β (IL-1β) cytokine release, thereby dampening innate immune responses. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
