Biological Activity

BBT-176 is a reversible, fourth-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor designed to target osimertinib-resistant mutations. It potently inhibits EGFR harboring C797S-containing triple and double resistance mutations, including Del19/T790M/C797S and L858R/T790M/C797S. By blocking EGFR phosphorylation, BBT-176 suppresses downstream MAPK/ERK and PI3K/AKT signaling pathways, thereby inhibiting cell proliferation and inducing apoptosis in resistant non-small cell lung cancer models. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
