Biological Activity

BBI-355 is an orally bioavailable, potent, and selective small-molecule inhibitor of checkpoint kinase 1 (CHK1). By disrupting CHK1 signaling in the DNA damage response pathway, it impairs replication stress management and abrogates cell cycle checkpoints in cancer cells with extrachromosomal DNA (ecDNA) oncogene amplifications. Consequently, BBI-355 induces DNA damage accumulation, suppresses tumor growth, and exhibits marked antitumor activity both as a monotherapy and combined with targeted agents. [1]

Solvent Solubility & Note
FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
