Biological Activity

BAY-293 is a selective inhibitor of the KRAS-SOS1 interaction with an IC50 of 21 nM. By binding to SOS1, it prevents SOS1-mediated guanine nucleotide exchange on KRAS, thereby blocking the formation of active GTP-bound KRAS. Consequently, BAY-293 suppresses downstream RAS signaling through the MAPK/ERK pathway and inhibits cell proliferation in KRAS-dependent cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
