BAY 2413555 is a selective positive allosteric modulator of the muscarinic acetylcholine receptor M2 (M2R). By potentiating endogenous acetylcholine signaling at M2R, it enhances parasympathetic tone to restore cardiac autonomic balance. Potentiation of M2R activation leads to G protein-mediated inhibition of adenylyl cyclase and suppression of cardiac sympathetic overdrive, making M2R positive allosteric modulation a promising therapeutic strategy for cardiovascular disorders. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
