Biological Activity

Empesertib (BAY1161909) is an orally bioavailable triazolopyridine derivative that functions as a potent and selective inhibitor of monopolar spindle 1 kinase. By inhibiting kinase activity, it impairs the spindle assembly checkpoint during mitosis, leading to premature mitotic exit, chromosome missegregation, and cell death. Consequently, disruption of this mitotic checkpoint machinery prevents proper cell division and exhibits antiproliferative activity against tumor cells. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
