Biological Activity

BAY-069 is a potent dual inhibitor of branched-chain amino acid transaminase 1 (BCAT1, IC50 = 31 nM) and BCAT2 (IC50 = 153 nM). By suppressing BCAT1/2-mediated transamination, the compound blocks the catabolism of essential branched-chain amino acids into keto acids and glutamate. This selective metabolic inhibition increases intracellular branched-chain amino acid accumulation in tumor cells and disrupts cancer-associated metabolic pathways without causing non-specific cytotoxicity.

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
