Biological Activity

Baricitinib phosphate is a selective inhibitor of JAK1 (IC50 5.9 nM) and JAK2 (IC50 5.7 nM), displaying ~70-fold selectivity over JAK3 and ~10-fold selectivity over Tyk2 without inhibiting c-Met or Chk2. By blocking JAK-STAT signaling pathways, it suppresses cytokine-driven intracellular signaling, downstream gene expression, and inflammatory mediator release, while also inhibiting viral entry into target host cells. A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
