Biological Activity

Barbadin is a selective β-arrestin/β2-adaptin interaction inhibitor, targeting β-arrestin 1 (IC50 19.1 μM) and β-arrestin 2 (IC50 15.6 μM). It selectively blocks agonist-induced endocytosis of β2-adrenergic, V2-vasopressin, and angiotensin II type-1 receptors without interfering with receptor-arrestin complex formation. Downstream, Barbadin blunts ERK1/2 signaling pathways and exhibits pro-apoptotic activity while inducing G0/G1 cell cycle arrest in agonist-stimulated cancer cells. [1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
