Biological Activity

Baloxavir is a first-in-class, potent cap-dependent endonuclease inhibitor that targets the PA subunit of the influenza virus polymerase complex. By selectively inhibiting cap-dependent endonuclease activity, baloxavir blocks viral RNA transcription and suppresses influenza viral replication in infected cells. This targeted inhibition disrupts the essential cap-snatching process, preventing viral mRNA synthesis and downstream replication across various influenza virus strains. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
