Biological Activity

BAL-0028 is an indazole-class inhibitor of NLRP3 activation (IC50 = 25 nM) that binds to the NACHT domain of NLRP3 with Kd values ranging from 104 to 123 nM. Blockade of the NLRP3 inflammasome pathway suppresses caspase-1-dependent maturation and secretion of pro-inflammatory cytokines, including IL-1β and IL-18. This downstream functional suppression prevents gasdermin D-mediated pyroptosis and exerts broad anti-inflammatory phenotypes.

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
