Biological Activity

AZD8797 (KAND567) is an orally active, allosteric non-competitive antagonist of the human chemokine receptor CX3CR1. By binding CX3CR1, it interferes with CX3CL1-induced G-protein activation and exhibits biased modulation of G-protein signaling and β-arrestin recruitment. This selective inhibition blocks CX3CL1-mediated leukocyte adhesion in human whole blood (IC50 300 nM) and cell migration, suppressing downstream neuroinflammatory and immune trafficking pathways. FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
