AZD5904 is a selective and irreversible inhibitor of human myeloperoxidase (MPO). Pharmacological inhibition of MPO by AZD5904 reduces 3-chlorotyrosine-modified cardiac myosin binding protein-C (MYBPC3) levels and restores MYBPC3 phosphorylation. Consequently, AZD5904 alleviates perturbed calcium signaling and corrects relaxation defects in hypertrophic cardiomyocytes, demonstrating that targeting MPO represents a promising therapeutic approach for improving myocardial relaxation and cardiac diastolic dysfunction. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
