Biological Activity

AZD4747 is a potent, selective, and blood-brain barrier-permeable inhibitor of mutant GTPase KRASG12C. By binding to mutant KRASG12C, it suppresses oncogenic RAS/MAPK signaling pathways, thereby inhibiting tumor cell growth, survival, and proliferation in KRASG12C-mutant malignancies. Additionally, its blood-brain barrier penetration allows for therapeutic brain exposure, making it capable of targeting central nervous system metastases arising from KRASG12C-driven primary tumors.
