Biological Activity

AZD9574 (Palacaparib) is a central nervous system-penetrant, selective inhibitor and DNA trapper of poly(ADP-ribose) polymerase 1 (PARP1). By selectively target-binding PARP1 over other PARP family isoforms and trapping PARP1 on damaged chromatin, AZD9574 blocks base excision repair mechanisms, leading to persistent single-strand and double-strand DNA breaks. This inhibition impairs DNA damage repair pathways and induces profound synthetic lethality and apoptotic cell death in homologous recombination-deficient tumor cells.[1][2]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
