AZ 3146 is a potent and selective monopolar spindle 1 (Mps1) kinase inhibitor with an IC50 of. Treatment with AZ 3146 impairs chromosome alignment and overrides the spindle assembly checkpoint (SAC) during mitosis. Inhibition of Mps1 by AZ 3146 prevents the kinetochore recruitment of key SAC components, including Mad1, Mad2, and centromere protein E (CENP-E), leading to accelerated mitotic exit. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
