Onalespib (AT13387) is a potent, selective heat shock protein 90 (Hsp90) inhibitor that binds Hsp90 with a Kd of 0.7 nM. In cellular assays, AT13387 demonstrates anti-proliferative activity across cancer cell lines, inhibiting A375 (IC50 18 nM), MV4-11 (IC50 12 nM), NCI-H1975 (IC50 22 nM), and SKBr3 (IC50 55 nM) growth. By destabilizing Hsp90 client proteins, it disrupts oncogenic signaling to induce apoptosis and suppress tumor cell proliferation. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
