Biological Activity

ASP2453 is a potent, selective, and covalent KRAS G12C inhibitor. It effectively disrupts the Son of Sevenless (SOS)-mediated interaction between KRAS G12C and Raf, suppressing downstream MAPK signaling pathways including ERK phosphorylation. Through this targeted inhibition, ASP2453 blocks cell proliferation and exhibits robust antitumor activity across KRAS G12C-mutated preclinical cancer models. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
