AS1708727 is an orally bioavailable Foxo1 inhibitor that potently suppresses G6Pase (EC50 0.33 μM) and PEPCK (EC50 0.59 μM) promoter activities. By blocking Foxo1-mediated transcriptional regulation, this compound downregulates critical gluconeogenic enzyme expression in hepatocytes. Consequently, AS1708727 inhibits hepatic gluconeogenesis, resulting in attenuated hepatic glucose production and improved glycemic regulation in diabetic disease models.
