Biological Activity

ART899 is a potent and specific allosteric inhibitor of DNA polymerase theta (Polθ), targeting its microhomology-mediated end joining (MMEJ) activity. By selectively impairing Polθ-mediated DNA damage repair during cell replication, ART899 disrupts downstream repair pathways and potently radiosensitizes tumor cells under both normoxic and hypoxic microenvironments. Consequently, ART899 enhances radiation efficacy, promoting sustained tumor growth inhibition and cell death in cancer models without causing radiosensitization in noncancerous tissues. [1]

FAQ
Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
