AQX-016A is a potent agonist of SH2 domain-containing inositol 5'-phosphatase 1 (SHIP1). By activating SHIP1, AQX-016A downregulates phosphoinositide 3-kinase (PI3K) and AKT signaling, suppressing immune cell activation and inflammatory responses. In vitro, AQX-016A induces apoptosis in cancer cell lines in a time- and dose-dependent manner. Furthermore, it inhibits DNFB-induced inflammation in mouse ear edema and cutaneous anaphylaxis models. Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
- Preheat media/saline to 37°C.
- Mix rapidly (pipette/vortex) upon addition.
- For complex mixes, always add the aqueous phase last.
- Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
