Biological Activity

APTO-253 is a small-molecule inducer of KLF4 that selectively inhibits c-Myc expression and upregulates CDKN1A (p21). By enhancing KLF4 transcriptional activity, APTO-253 modulates the p21/c-Myc axis to suppress oncogenic signalling in acute myeloid leukemia cells. This regulatory response leads to G0-G1 cell-cycle arrest and triggers apoptotic cell death, thereby halting proliferation in leukemic cell populations. [1]

Q: Why does a DMSO stock precipitate in media/saline?
A: It is caused by unheated aqueous media (supersaturation), local solvent shock, or incorrect mixing sequence.
How to fix it?
  • Preheat media/saline to 37°C.
  • Mix rapidly (pipette/vortex) upon addition.
  • For complex mixes, always add the aqueous phase last.
  • Re-dissolve minor precipitates with a 37°C water bath and sonication (≤30 min).
